VistaGen Therapeutics, Inc. (VSTA) Announces Identification of Cell Surface Marker that Permits High-Yield Purification of Human Pluripotent Stem Cell-Derived Cardiomyocytes

VistaGen Therapeutics, Inc., a biotechnology company applying stem cell technology for drug rescue and cell therapy, just announced the publication of original research funded by the Company that identifies an antibody useful in the identification and purification of cardiac progenitors and cardiomyocytes.

The research, titled “Novel Use for SIRPA as a Specific Cell Surface Marker for the Isolation of Human Pluripotent Stem Cell-Derived Cardiomyocytes,” stems from research funded in part by VistaGen and conducted by a collaborative team led by Dr. Gordon Keller at the University Health Network’s McEwen Centre for Regenerative Medicine in Toronto. The results of these studies were published in the peer-reviewed journal, Nature Biotechnology, on October 23, 2011.

“The identification and use of the SIRPA antibody permits us to select the very earliest human cardiac progenitors, as well as mature cardiomyocytes, and study the important role of non-cardiomyocytes in the function and maturation of cardiomyocytes,” said Dr. Ralph Snodgrass, President and Chief Scientific Officer of VistaGen. “Using this antibody, we can produce human cardiomyocytes with greater than 95% purity, without genetically modifying the cells and without antibiotic selection, which is a significant step forward for our cardiotoxicity bioassay system, CardioSafe 3D™, and our cell therapy initiatives.”

The Keller team identified human cardiomyocyte specific markers by screening human embryonic stem cell (hESC)-derived cardiovascular populations with known antibodies. From this screen, the signal-regulatory protein alpha (SIRPĪ‘) was identified as a marker expressed specifically on hESC and induced pluripotent stem cell (iPSC)-derived cardiomyocytes and their precursors. Cell sorting and selection with the SIRPA antibody allowed for the enrichment of cardiac precursors and cardiomyocytes from hESC/iPSC differentiation cultures, yielding populations of up to 98% cardiac troponin T-positive cells. SIRPA-positive cells, when cultured, express the expected markers, transcription factors and cytoskeletal markers of cardiomyocytes, and can be maintained in culture for extended periods of time. These findings provide, for the first time, a simple method for isolating some of the earliest populations of cardiac precursors and mature cardiomyocytes from human pluripotent stem cell cultures. This readily adaptable technology offers a viable approach for generating large numbers of enriched, non-genetically modified, cardiomyocytes for numerous therapeutic applications.

Through its long-standing collaboration with Dr. Gordon Keller, who is also Chairman of the Company’s Scientific Advisory Board, and a license agreement with the University Health Network, VistaGen has exclusive worldwide rights to intellectual property arising from this research conducted by Dr. Keller’s laboratory. These studies, funded by VistaGen, are part of the Company’s Human Clinical Trials in a Test Tubetm platform, which has proprietary applications in drug screening, cell therapy, and regenerative medicine.

Mainland Resources, Inc. (MNLU) Announces Addition of One of World’s Best Known Petroleum Geologists to Advisory Board

Mainland Resources, Inc., an independent oil and gas exploration, development and production company, today announced the addition of Dr. George V. Chilingar, PhD to its Advisory Board effective immediately. Dr. Chilingar will assist the company with a strategic partnership alliance and the supporting financing for their Buena Vista shale gas project in Mississippi. Dr. Chilingar is one of the world’s best known petroleum geologists and has extensive knowledge and experience in the oil and gas industry. He has been recognized for the discovery and extraction of oil reserves around the world.

Dr. Chilingar has been honored by several nations for his works in the field of the geology of petroleum reservoirs, petrophysics, sedimentology and petroleum engineering, including India, Russia, Saudi Arabia and Iran. One of his greatest contributions to the petroleum industry is his method for identifying oil-rich rock by analyzing the ratio of calcium to magnesium in core samples, which helped discover one of Iran’s largest oil fields.

“I am excited by the potential of Mainland’s Buena Vista shale gas project in Mississippi. It is a significant prospect where I feel I can assist the Company both with strategic partnership affiliation, financing and a greater understanding of the long term development of this play,” said Dr. Chilingar in a press release on Monday.

Mainland’s Mississippi prospect is located along the Gulf Coast Salt basin in a region where Mainland believes there is an extension of the Haynesville shale similar to areas in Louisiana that have gained notoriety for recent discoveries. Their proprietary data on previous drilling of a deep well makes them convinced of its potential for major shale gas production. Mainland Resources had previously announced plans to merge with American Exploration which would give Mainland 92 percent of the 28 sections in the Buena Vista prospect which could be up to 500 BCF/section based on the cored interval but as of yet the deal has not be done.

“We are extremely pleased to have Dr. Chilingar join our Advisory Board to assist with a strategic partnership alliance. His world-class expertise lends tremendous credibility to our current Mississippi project and shows the caliber of talent that our Company is attracting. His insight and contribution will be greatly appreciated by our management,” said Mike Newport, President, Mainland Resources in Monday’s press release.

Aeolus Pharmaceuticals, Inc. (AOLS) Receives Multi-year Award to Develop Treatment against Nerve Agent Exposure

Aeolus Pharmaceuticals, Inc., a biotechnology company developing a novel platform of compounds to treat radiation and chemical exposure, as well as oncology indications, today announced that NIH CounterACT has awarded a $735,951 contract to Manisha Patel, Ph.D., at the University of Colorado Anschutz Medical Campus. The funding is to support the development of AEOL 10150, Aeolus’ broad-spectrum catalytic antioxidant, as a medical countermeasure (MCM) against nerve agents.

The grant was awarded with primary objectives in mind, including the evaluation of the neuroprotective efficacy of AEOL 10150 against pilocarpine (an organic compound of plant origin)-induced neurotoxicity in rat models; exposure to nerve agents, metabolic poisons, or high levels of sulfur mustard can cause seizures and unconsciousness.

Aeolus said the two-year award builds on research conducted by Dr. Patel in rodent models of neurotoxicity and neurodegeneration. Dr. Patel’s studies have demonstrated that oxidative stress plays a central role in seizure-induced brain injury. Oxidative stress plays a key role in seizure-induced brain injury. Since nerve agents provoke seizures, it is vital to determine AEOL 10150’s neuroprotective abilities against such agents.

“We are pleased to see the expansion of the research and development of AEOL 10150 as a broad spectrum medical countermeasure, and are grateful for the support that NIH CounterACT provides for the development of this promising compound. With this project, the U.S. Government is now funding research and testing of AEOL 10150 in five MCM indications,” John L. McManus, president and CEO of Aeolus stated in the press release.

Aeolus noted that previous studies have also shown AEOL 10150 as an effective countermeasure to protect the lungs from damage due to inhalation of chlorine gas and sulfur mustard gas. The compound is currently being developed for a wide variety of applicable medical countermeasures with support from the U.S. Government.

Product Funding Review:

NIH CounterACT is funding research and development of the compound as a countermeasure against exposure to nerve agents, chlorine gas and sulfur mustard gas. NIH-NIAID is conducting animal efficacy studies of the compound as a countermeasure for the Gastro-Intestinal (GI) effects of Acute Radiation Syndrome (ARS); and the Biomedical Advanced Research and Development Authority (BARDA) has awarded the Aeolus a contract valued up to $118 million to develop AEOL 10150 as a countermeasure against Lung ARS/Delayed Effects of Radiation Exposure (DEARE).

GLG Life Tech Corp. (GLGL) Announces 2011 Agriculture R&D Program Results

GLG Life Tech Corp., a company that engages in growing, refining, producing, and distributing stevia extract to the food and beverage industry worldwide, has announced the results of its 2011 Agriculture R&D Program.

The Company reported that it has successfully reached another milestone in its stevia seed propagation program and that its Huinong Three (H3) proprietary stevia varietal propagation is on schedule. It has been confirmed that H3 will be available for planting in the growing season of 2012. These new seeds will provide GLG with a substantial cost reduction in its production of high-purity stevia extracts. This is because of both the very high levels of rebaudioside A (“RA”) that is naturally found in these plants and the higher leaf yields produced from the larger H3 plants. The H3 plants have about 76% RA in the plant leaf, which is 26% higher than the first generation (H1) seeds. Also, the H3 plants will generate 46% more leaf per acre than was produced by the earlier H1 plants. RA is the sweetest component of the stevia plant and the primary glycoside used to meet with the demand for stevia sweeteners in the market.

GLG is also announcing the next generation of plant verietals. The Huinong Four (H4) is on schedule to be commercially available to be distributed to its contract farmers for the 2012 stevia growing season. The H4 proprietary strain displays a 16% increase in leaf yield over the H3 plants, while keeping a similar 76% RA content. This will result in more reductions to overall production costs. The H4 seeds will be playing a vital role, as parents, in the advance of the Company’s next generation of seeds.

The GLG Agriculture R&D team has also created a new Huinong Five (H5) plant strain this year, which is able to produce high amounts of stevioside (STV). The H5 strain’s STV content of about 70% makes it’s the highest STV seed that is available in the world today, with a leaf yield between the H2 and H3 yields. H5 will aid GLG in further reducing production costs for its BlendSure™ products, which has already received GRAS certification and a Letter of No Objection from the FDA earlier on this year.

GLG Chief Agricultural Scientist Mr. Qibin Wang stated, “I am very pleased with the recent developments of our agricultural R&D program. Under Dr. Luke Zhang’s leadership, we have progressively achieved several very significant milestones, with H4 and H5 following on the heels of the H1, H2 and H3 proprietary stevia plant varietals. We will continue to innovate agriculturally and to develop non-GMO, high yielding stevia strains that are natural, sustainable, and provide value for our future growth.”

GLG Chairman and CEO Dr. Luke Zhang commented, “GLG remains in the forefront of the stevia industry with these important agricultural achievements. Advancements such as these underline the importance of our vertical integration from seed to shelf and our control over the stevia supply chain. Leaf costs account for approximately 65% of the cost to produce high purity stevia extracts so our agriculture advances have the most significant impact on reducing our product costs. We expect that global stevia demand will continue to increase in the future as the cost of stevia extracts decreases, and flavor and aftertaste challenges are overcome. GLG has all the tools to address taste and cost challenges that have impacted stevia’s adoption in the past. We are confident that stevia demand from the global market will increase now that the aftertaste issue has been resolved by our recently established AN0C Stevia Solutions Company, comprised of stevia and Food & Beverage formulation specialists who work together and have a deep understanding in both stevia and product formulations. We expect to continue to lower our product costs through our dedicated agricultural R&D program. GLG is in the leading position to provide our customers both good-tasting stevia solutions and low cost stevia-based sweeteners.”
